چکیده

A novel extension to a previous method has been proposed to evaluate the nature of dry powder mixing processes using a mixing sensitive colouring agent. The method proposed measures the change in the colour (hue) and colour intensity of the powder blends over mixing time. To evaluate the extent of fine particle dispersion and de-agglomeration, cohesive and free flowing lactose (white) and cohesive micronized iron oxide (dark red) were blended as a bulk and tracer respectively. Measuring the change in hue and intensity of micronized iron oxide in a lactose bulk enables the analysis of two distinct mixing behaviours, namely the deagglomeration of the pigment (transition from red to orange) and the dispersion of a given sized aggregate through the bulk material (increase in the intensity of the blend’s hue). Blends were manufactured using two different types of powder mixers at a range of processing conditions. Samples were withdrawn from the mixers at regular time intervals and the hue and intensity analysed using a colourimeter. Hue values were assembled to create a series of process specific curves. The colourimetry data was able to clearly separate and group the mixers into low and high levels of shear and/or mixing intensity over a range of conditions, and suggests that the gradual dispersion and de-agglomeration can be monitored independently using this method. These initial results suggest that the iron oxide tracer method could provide the basis for a novel quantitative approach to producing equivalent powder blends as a function of mixer type, scale and operating conditions.

Extrinsic lactose fines improve dry powder inhaler formulation performance of a cohesive batch of budesonide via agglomerate formation and consequential co-deposition.

The aim of the study was to investigate how the fine particle content of lactose carriers prepared with different types of lactose fines regulates dry powder inhaler (DPI) formulation performance of a cohesive batch of micronised budesonide. Budesonide formulations (0.8 wt%) were prepared with three different lactose carriers (Lactohale (LH) LH100, 20 wt% LH210 in LH100 and 20 wt% LH300 in LH10...

Evaluation of the degree of mixing of combinations of dry syrup, powder, and fine granule products in consideration of particle size distribution using near infrared spectrometry.

We used near infrared (NIR) spectroscopy to evaluate the degree of mixing of blended dry syrup (DS) products whose particle sizes are not specified in the Revised 16th Edition of the Japanese Pharmacopoeia, and also evaluated the degree of mixing when powder products or fine granule products were added to DS products. The data obtained were used to investigate the relationship between the parti...

Sustained Release Multiparticulates via Powder Coating

Hydrophobic grade fumed silicas are generally used in tabletting as lubricants. They are also found to retard the release of pharmaceutical actives. This is because of their hydrophobic nature, which gives good cohesive and adhesive properties. considering this point it was thought that they could be used as dry coating agents for solid dosages forms. The model drug selected for this work was p...

DEM Analysis of Particle Adhesion during Powder Mixing for Dry Powder Inhaler Formulation Development

Understanding the adhesive interactions between active pharmaceutical ingredient (API) particles and carrier particles in dry powder inhalers (DPIs) is critical for the development of formulations and process design. In the current study, a discrete element method (DEM), which accounts for particle adhesion, is employed to investigate the attachment processes in DPIs. A critical velocity criter...
